%X Actinoporins are soluble proteins capable of forming pores in biological membranes containing sphingomyelin (SM). Among them, sticholysin I (Stn I), a toxin produced by Stichodactyla helianthus, has attracted attention because of its potential senolytic activity. Given the fundamental role of lipids in the recognition and activity of these proteins and the fact that senescent cells expose a higher proportion of phosphatidylserine (PS) on the outer leaflet of their plasma membrane, the aim of this work was to investigate the influence of PS on the interaction of Stn I with model membranes. To this end, recombinant Stn I was produced and purified from Escherichia coli, and its structural characterization confirmed that the purified protein retained its native conformation. Subsequently, its interaction with DOPC:SM:DOPS lipid vesicles was studied by isothermal titration calorimetry (ITC) and calcein leakage assays to evaluate membrane-binding affinity and pore-forming activity, respectively. The results showed that the incorporation of PS at the proportions studied did not significantly affect either of these properties, suggesting that other factors may contribute to the reported selectivity of Stn I toward senescent cells. %~
