    El patrimonio veterinario como herramienta didáctica: elaboración de material multimedia por estudiantes de Veterinaria y Bellas Artes para la docencia de la Historia de la Veterinaria y la difusión del Museo Veterinario Complutense

    Los profesores de Historia de la Veterinaria vienen presentando y desarrollando desde el curso 2011 sucesivos proyectos de innovación que han permitido dinamizar la enseñanza de esta materia interdisciplinar, entre humanidades y ciencias. Como continuación de esta trayectoria y para completar los resultados de los anteriores proyectos se propone incluir en la innovación docente el patrimonio disponible en el Museo Veterinario Complutense. A partir de su amplia y variada gama de piezas se persigue innovar la docencia de la asignatura a través de material multimedia elaborado por estudiantes. La propuesta se centra en el trabajo colaborativo, el desarrollo del concepto aula invertida, la interacción entre centros y estudiantes de facultades UCM (Veterinaria y Bellas Artes) y de otros países (internacionalización). Para ello, profesores de Veterinaria de diversos departamentos (especializados en Historia de la veterinaria) y de Bellas Artes (especializados en patrimonio e imagen) coordinarán a estudiantes de ambos centros con la colaboración de PAS con experiencia en la toma de imágenes y grabación y PAS de la biblioteca de Veterinaria. Se elaborará a partir de las piezas un documento y un material audiovisual. Éste a su vez lo deben difundir por redes y en presentaciones con posterior debate a otros estudiantes de sus centros y de otras facultades de diferentes países para completar contenidos y ampliar la difusión (incluyendo centros de Sudamérica coordinados por la facultad Veterinaria de Litoral, Argentina). En el aspecto docente el material se incluirá en el campus virtual como material didáctico para la asignatura Cultura, Historia e Identidad Veterinaria. Para su ejecución se harán equipos siguiendo tres apartados dentro de la evolución de la veterinaria: medicina y terapéutica animal, producciones animales y la seguridad alimentaria. Se introducirán en los textos contenidos alusivos a conflictos interculturales, temas de género e inclusión. Como ejemplo con el material de herrado y forja, testimonio de una antigua asignatura obligatoria y limitante, se explicará la difícil incorporación de la mujer en veterinaria. Asimismo, se hará una versión especial en inglés para su difusión a centros extranjeros y visitas de museo de otros países. Los materiales que se obtengan completarán y enriquecerán el repositorio digital de contenidos de la asignatura y los resultados del proyecto se difundirán a través de asociaciones científicas, congresos de docencia, la web de la facultad de Veterinaria y a través del Museo Veterinario.

    PK/PD Analysis by Nonlinear Mixed-Effects Modeling of a Marbofloxacin Dose Regimen for Treatment of Goat Mastitis Produced by Coagulase-Negative Staphylococci

    Coagulase-negative staphylococci are main pathogens that produce goat mastitis. Marbofloxacin is a third-generation fluoroquinolone approved for treat mastitis in animals. The objectives of this study were: (i) to determine the pharmacokinetics of marbofloxacin (10 mg/kg/24 h) in serum and milk administered intramuscularly for five days in goats with mastitis induced by coagulase-negative staphylococci; (ii) to characterize the concentration–effect relationship of marbofloxacin against coagulase-negative staphylococci in Mueller Hinton broth and goat milk; (iii) to determine AUC/MIC cutoff values of marbofloxacin, and (iv) to perform a PK/PD analysis to evaluate the efficacy of the dose regimen for the treatment of goat mastitis produced by coagulase-negative staphylococci. Marbofloxacin presented context-sensitive pharmacokinetics, influenced by the evolution of the disease, which decreased marbofloxacin disposition in serum and milk. Marbofloxacin showed a median (95% CI) fAUC/MIC values for MIC of 0.4 and 0.8 µg/mL of 26.66 (22.26–36.64) and 32.28 (26.57–48.35) related with −2 log10CFU/mL reduction; and 32.26 (24.81–81.50) and 41.39 (29.38–128.01) for −3 log10CFU/mL reduction in Mueller Hinton broth. For milk, −2 log10CFU/mL reduction was achieved with 41.48 (35.29–58.73) and 51.91 (39.09–131.63), and −3 log10CFU/mL reduction with 51.04 (41.6–82.1) and 65.65 (46.68–210.16). The proposed dose regimen was adequate for the treatment of goat mastitis produced by coagulase-negative staphylococci, resulting in microbiological and clinical cure of all animals. The animal model used in this study provided important pharmacokinetic information about the effect of the infection on the pharmacokinetics of marbofloxacin. Pharmacodynamic modeling showed that fAUC/MIC cutoff values were higher in goat milk compared with Mueller Hinton broth.

    Desde un enfoque “one health”, la salud de los animales se refleja en la salud pública y la salud medioambiental. ¿La buena praxis de los profesionales responsables de la salud animal, se refleja en el bienestar emocional de estos profesionales sanitarios?. La OMS define el bienestar emocional como ese estado de ánimo en el cual la persona se da cuenta de sus propias aptitudes, puede afrontar las presiones normales de la vida, trabajar productivamente y contribuir a la comunidad. El estudio WSAVA 2019, sobre bienestar veterinario, ansiedad y depresión, muestra que la salud mental de los profesionales más jóvenes está seriamente en riesgo. EL estrés laboral ha sido catalogado por la OMS como la epidemia del siglo XXI. El desarrollo del programa atención consciente y gestión emocional, en el marco académico en el que se integra, se alinea con los objetivos de desarrollo sostenible de la ONU (Agenda 2030) ODS3 y transversalmente con ODS5 y ODS16.

    Coffee Silverskin Extract: Nutritional Value, Safety and Effect on Key Biological Functions

    This study aimed to complete the scientific basis for the validation of a coffee silverskin extract (CSE) as a novel food ingredient according to European legislation. Nutritional value, safety, effects on biochemical biomarkers and excretion of short chain fatty acids (SCFAs) in vivo of CSE were assessed. Proteins, amino acids, fat, fatty acids, fiber, simple sugars and micronutrients were analyzed. For the first time, toxicological and physiological effects were evaluated in vivo by a repeated-dose study in healthy Wistar rats. Hormone secretion, antioxidant (enzymatic and no-enzymatic) and anti-inflammatory biomarkers, and dietary fiber fermentability of CSE (analysis of SCFAs in feces) were studied in biological samples. This unique research confirms the feasibility of CSE as a human dietary supplement with several nutrition claims: “source of proteins (16%), potassium, magnesium, calcium and vitamin C, low in fat (0.44%) and high in fiber (22%)”. This is the first report demonstrating that its oral administration (1 g/kg) for 28 days is innocuous. Hormone secretion, antioxidant or anti-inflammatory biomarkers were not affected in heathy animals. Total SCFAs derived from CSE fiber fermentation were significantly higher (p < 0.05) in male treated rats compared to male control rats. All the new information pinpoints CSE as a natural, sustainable and safe food ingredient containing fermentable fiber able to produce SCFAs with beneficial effects on gut microbiota.

    Effect of chemically modified tetracycline-8 (CMT-8) on hematology, blood chemistry, cytokines and peripheral blood lymphocyte subsets of healthy dogs

    Tetracyclines are antibiotics widely used in human and veterinary medicine. Effects on the immune system and inflammatory response, including effects on blood leukocytes proliferation and function and in cytokines synthesis, have been described. Chemically modified tetracyclines (CMT) have lost their antimicrobial activity, but maintain these other properties. This study analyzes the effect of chemically modified tetracycline-8 (CMT-8) on the evolution of complete blood count, blood chemistry, the mRNA expression of selected cytokines and peripheral blood lymphocyte subpopulations distribution in healthy dogs. CMT-8 at a dose of 10 mg/kg once daily was administered per os to six healthy dogs. A control group of five healthy dogs, living in the same conditions than dogs treated with CMT-8, received placebo with an identical therapeutic regimen. When given at the doses used in this study, no side effects of CMT-8 were detected, suggesting a good tolerance and a limited toxicity of the drug. Dogs treated with CMT-8 showed a gradual increase in mean corpuscular hemoglobin. The administration of CMT-8 in healthy dogs did not affect blood mRNA expression of IFN-γ, TNFα, IL-4, IL-6, IL-10, IL-12 p40 and IL-13. However, the lymphocytes expressing class II MHC on their surface decreased during the first two weeks of CMT-8 treatment and subsequently increased for the next three months. Considering the absence of antimicrobial properties of the drug, the effects of CMT-8 detected in this study seem to be unrelated to the classical antimicrobial activity attributed to tetracyclines.

    Bioaccesibility, Metabolism, and Excretion of Lipids Composing Spent Coffee Grounds

    The bioaccessibility, metabolism, and excretion of lipids composing spent coffee grounds (SCGs) were investigated. An analysis of mycotoxins and an acute toxicity study in rats were performed for safety evaluation. Total fat, fatty acids, and diterpenes (cafestol and kahweol) were determined in SCGs and their digests obtained in vitro. A pilot repeated intake study was carried out in Wistar rats using a dose of 1 g SCGs/kg b.w. for 28 days. Fat metabolism was evaluated by analysis of total fat, cholesterol, and histology in liver. The dietary fiber effect of SCGs was measured radiographically. The absence of mycotoxins and toxicity was reported in SCGs. A total of 77% of unsaturated fatty acids and low amounts of kahweol (7.09 µg/g) and cafestol (414.39 µg/g) were bioaccessible after in vitro digestion. A significantly lower (p < 0.1) accumulation of lipids in the liver and a higher excretion of these in feces was found in rats treated with SCGs for 28 days. No lipid droplets or liver damage were observed by histology. SCGs acutely accelerated intestinal motility in rats. SCGs might be considered a sustainable, safe, and healthy food ingredient with potential for preventing hepatic steatosis due to their effect as dietary fiber with a high fat-holding capacity.

    Validation of coffee by-products as novel food ingredients

    This research aimed to validate coffee husk, parchment and silverskin as new health-promoting food ingredients. Characterization of the novel ingredients was carried out by Raman and infrared spectroscopy and analysis of total phenolic compounds, chlorogenic acid, caffeine and dietary fiber. Antioxidant properties of the novel ingredients were tested by ABTS and intracellular ROS formation in HepG2 cells. Pesticides, mycotoxins, acrylamide and acute toxicity experiments following OECD Test Guidelines 425 were performed to assess the food safety of extracts, solid residues and raw materials. Husk and silverskin are proposed as a source of two food ingredients: aqueous extracts enriched in phytochemicals and antioxidant dietary fiber while parchment is proposed as a natural source of antioxidant dietary fiber. No lesions were found in selected isolated vital organs from treated animals. Coffee by-products can be converted into safe food ingredients allowing a whole food waste recovery. Analyses of contaminants are essential for achieving this goal.

    Pharmacokinetic Behaviour of Enrofloxacin after Single Intramuscular Dosage in American Black Vultures (Coragyps atratus)

    The aim of the study was to investigate the intramuscular pharmacokinetics of enrofloxacin in black vultures (Coragyps atratus). The pharmacokinetics of a single intramuscular dose (10 mg/kg) of enrofloxacin was studied in six vultures. Plasma concentrations of enrofloxacin and its active metabolite, ciprofloxacin, were determined by high-performance liquid chromatography (HPLCuv). Pharmacokinetic parameters were estimated using non-compartmental and compartmental analysis. After intramuscular administration, enrofloxacin showed a rapid and complete absorption, reaching a Cmax value of 3.26 ± 0.23 μg/mL at 1.75 ± 0.53 h. A long terminal half-life of 19.58 h has been observed. Using previously published MIC values to perform a PK/PD analysis, cumulative fraction responses obtained after Monte Carlo simulation for AUC/MIC > 30, 50 and 125 were 72.93%, 72.34% and 30.86% for E. coli and 89.29%, 88.89% and 58.57% for Mycoplasma synoviae, respectively. Cumulative fraction responses obtained for Cmax/MIC index were 33.93% and 40.18% for E. coli and M. synoviae, respectively. The intramuscular administration of 10 mg/kg could be appropriate to treat infectious diseases caused by gram-positive bacteria with MIC value lower than 1 µg/mL; however, although enrofloxacin showed a slow elimination in black vultures, plasma concentrations were insufficient to reach the gram-negative stablished breakpoints.

    Proyecto de innovación docente enfocado en valores nucleares y convivencia en una sociedad serodiscordante, es continuidad del P89/2018, titulado “competencias intra e interpersonales, sexo consciente, fármacos y mindfulness”. Durante el primer semestre, desde el paradigma de aprendizaje cooperativo, basado en resolución de proyectos, se desarrolló la investigación documental, reflexión y debate sobre estos centros de interés: valores nucleares & VIH: abordaje de la seropositividad; adherencia terapéutica; estudio PARTNER indetectable es igual a intransmisible; profilaxis pre-exposición (PrEP); convivir en serodiscordancia; encuestas sobre estigmatización y discriminación social y sanitaria; riesgos globales de la discriminación terapéutica; relaciones interpersonales y emociones aflictivas; mindfulness y compasión basado en la evidencia; psicología positiva y resiliencia. En el segundo semestre, durante la fase cero del estado de alerta sanitaria, se desarrolló una intervención psicoeducativa, dirigida a facilitar estrategias que ayudasen a gestionar mejor el impacto emocional generado por la incertidumbre, la restricción de movilidad territorial, el confinamiento, y el miedo al contagio de la COVID-19. Perpetuar el estigma ante VIH, es una violación de los derechos humanos. Como antídoto a la indiferencia, se integró en nuestra intervención el fomento de la compasión activa, entendida como un sentido básico de cuidado, sensibilidad y apertura hacia el sufrimiento propio y de los demás, y la intención genuina de intentar aliviarlo y prevenirlo. La pandemia generada por el SARS-CoV-2 es una oportunidad para entender que ante este virus, aquí y ahora, la única opción es prevenirlo, detectarlo mediante pruebas y confinarlo, es decir no transmitirlo y no reinfectarse. Si de forma transversal somos capaces de interiorizarlo y normalizar la convivencia en serodiscordancia, si normalizamos las pruebas diagnósticas rutinarias, podremos entre todas y todos reducir hasta un 90% la transmisión activa otra pandemia de la que hoy no se habla, del VIH, de este modo conseguiríamos minimizar la aparición de nuevos casos, frenar su avance y vencer definitivamente al SIDA.
