Abstract
Legionella pneumophila is ubiquitous in aquatic environments, invading protozoa and causing pneumonia in humans. The L. pneumophila genome encodes a multigene family of 11 ankyrin (ank) genes. Isogenic mutants indicate AnkH and AnkJ are involved in robust intracellular replication in protozoa (Acanthamoeba polyphaga, Hartmanella vermiformis, Tetrahymena pyriformis) and human macrophages, with upregulation mediated by RpoS. The proteins’ modularity suggests a role in phagosome biogenesis and adaptation to diverse hosts. Thus, co-evolution with protozoa may have enabled L. pneumophila to parasitize humans.
