Revisión de la terapia combinada con donepezilo y memantina en el tratamiento de la enfermedad de Alzheimer

Abstract

La destrucción neuronal y posterior retracción del encéfalo son el diagnóstico más concluyente de la presencia de la enfermedad de Alzheimer. Como ya conocemos, la enfermedad tiene dos rasgos microscópicos característicos que son la existencia de depósitos extracelulares de proteína β-amieloide y los ovillos neurofibrilares intraneuronales (constituido por la conocida proteína Tau), ambos en forma de plegamiento anómalo. Actualmente existe prescripción de tratamientos con donepezilo y memantina de forma combinada. Se han hecho muchos estudios para descubrir la efectividad, las diferencias con la monoterapia, la seguridad y tolerancia. Mi trabajo se ha dirigido a revisar estudios seleccionados, buscando esclarecer estos puntos citados anteriormente, pudiendo concluir que en la mayoría de los ensayos realizados existe un beneficio significativo o modesto de la terapia combinada tanto a nivel cognitivo, de comportamiento y en la función y estado global del paciente.

Neuronal destruction and subsequent retraction of the brain are the most conclusive diagnosis of the presence of Alzheimer's disease. As we know, the disease has two characteristic microscopic features that are the existence of extracellular deposits of βamyloid protein and intraneuronal neurofibrillary tangles (known protein consisting Tau), both as misfolding. There currently prescribed treatments donepezil and memantine in combination. There have been many studies to discover the effectiveness, differences with monotherapy, security and tolerance. My work has been directed to review selected studies, seeking to clarify these points mentioned above, concluding that most trials there is a significant or modest benefit of combination therapy both cognitive, behavioral and the function and status overall patient.
